Transcription-initiated DNA Replication in Yeast

 

DNA polymerases begin DNA synthesis by addition of nucleotides to primase-generated RNA molecules. Transcription-dependent RNA:DNA hybrids may also have the potential to initiate replication, although such events has never been observed in eukaryotic cells. To promote transcription-dependent replication, we generated RNA:DNA hybrid promoting conditions by inhibition of Toposiomerase 1 in the absence of RNaseH activities. As a consequence, we detected hitherto unknown replication events in ribosomal DNA that we named transcription-initiated replication (TIR) because they depend on the transcriptional activity of RNA polymerase I. I will present data on factors and mechanisms involved in TIR. Our results suggest a link between torsional stress in highly trasncribed genes, RNA:DNA hybrids, non-canonical DNA replication and chromosomal alterations.
